碱性/中性转化酶(NINV)催化蔗糖分解成葡萄糖和果糖,是木薯块根的蔗糖分解代谢的关键酶之一。本研究以淀粉含量不同的木薯品种(SC8-高淀粉,SC124-中淀粉,9Ⅰ-低淀粉)为材料,研究木薯块根膨大期,Me NINV基因家族在源库器官的表达模式以及酶活性特点。结果表明,不同品种木薯Me NINVs的表达模式相似,Me NINV1、Me NINV16、Me NINV110及n INV1在叶片中表达量均高于其他成员;同时,Me NINV1和NINV1在块根中的表达量也高于其他成员。不同品种木薯叶片中的Me NINVs表达及酶活性差别不大,但是高淀粉品种SC8块根中的Me NINVs表达量比9Ⅰ低,而NINV酶活性却远高于9Ⅰ,推测木薯NINV酶活性存在转录后调控机制。本研究为进一步研究碱性/中性转化酶在木薯淀粉积累过程中的作用奠定了基础。
Alkaline / neutral invertase (NINV) catalyzes the breakdown of sucrose into glucose and fructose, which is one of the key enzymes of sucrose catabolism in cassava roots. In this study, the cassava varieties (SC8-high starch, SC124-medium starch, 9Ⅰ-low starch) with different starch content were used to study the expression pattern of Me NINV gene family and enzyme activity in the tuber expansion stage. The results showed that Me NINV1, Me NINV16, Me NINV110 and n INV1 were all expressed higher in leaves than Me NINV1. The expression levels of Me NINV1 and NINV1 in tuberous roots were also higher than those of Me NINV1, other members. Me NINVs expression and enzyme activities in different varieties of cassava leaves were not significantly different, however, the expression level of Me NINVs in high starch varieties SC8 was lower than that of 9Ⅰ, while NINV activity was much higher than that of 9Ⅰ. Regulatory mechanism. This study laid the foundation for the further study on the role of alkaline / neutral invertase in the accumulation of tapioca starch.
