目的探讨PIAS3过表达对人脑胶质瘤U251细胞生长的作用及其可能的机制。方法通过实时荧光定量PCR检测PIAS3在正常脑细胞和脑胶质瘤U25 1细胞中的表达情况。转染PIAS3过表达载体,提高U251细胞中PIAS3表达水平,通过噻唑蓝(MTT)试验检测细胞增殖情况。Western Blot验证PIAS3表达与增殖相关基因PI3K及Akt间关系。结果 PIAS3在正常脑组织中相对高表达,而在人脑胶质瘤U251细胞中表达明显下降(P<0.01)。体外转染PIAS3过表达载体能明显抑制U251细胞生长,并且有效抑制PI3K活性及p-Akt表达,但对总的Akt无明显影响。结论胶质瘤U251细胞中异常低表达的PIAS3发挥着重要的增殖促进作用,过表达PIAS3可通过抑制PI3K/Akt通路有效抑制U251细胞增殖。
Objective To investigate the effect of PIAS3 overexpression on the growth of human glioma U251 cells and its possible mechanism. Methods The expression of PIAS3 in normal brain cells and glioma U25 1 cells was detected by real-time fluorescence quantitative PCR. The PIAS3 overexpression vector was transfected to enhance the expression of PIAS3 in U251 cells. The cell proliferation was detected by MTT assay. Western Blot to verify PIAS3 expression and proliferation-related genes PI3K and Akt relationship. Results PIAS3 was relatively higher in normal brain tissue and decreased in human glioma U251 cells (P <0.01). In vitro transfection PIAS3 overexpression vector can significantly inhibit the growth of U251 cells, and effectively inhibit PI3K activity and p-Akt expression, but no significant effect on the total Akt. Conclusions PIAS3, an abnormally low expression of glioma U251 cells, plays an important role in proliferation. Overexpression of PIAS3 can effectively inhibit the proliferation of U251 cells by inhibiting PI3K / Akt pathway.
