目的研究细胞S期激酶相关蛋白SKP2(S-PHASE KINASE-ASSOCIATED PROTEIN2)在食管癌组织中的表达及其临床意义。方法应用免疫组化S-P方法检测121例食管癌组织中SKP2蛋白与P27蛋白的表达水平和90例正常食管组织中的SKP2蛋白表达水平。结果SKP2蛋白表达率在食管癌中为64.4%(78/121),而正常食管组织中为13.3%(12/90),差异有显著性(P<0.05)。SKP2蛋白阳性表达率与患者年龄、性别、肿瘤大小、有无淋巴结转移及TNM分期无关(P>0.05),与肿瘤分化程度有关(P<0.05)。SKP2与P27表达呈显著负相关(P<0.05)。结论从蛋白水平证明SKP2异常表达可促进食管癌的发生发展,并将可能为食管癌的早期诊断和治疗提供新的手段。
Objective To study the expression of S-PHASE KINASE-ASSOCIATED PROTEIN 2 (S-PHASE KINASE-ASSOCIATED PROTEIN2) in esophageal carcinoma and its clinical significance. Methods The expression of SKP2 protein and P27 protein in 121 cases of esophageal carcinoma tissue and the expression of SKP2 protein in 90 cases of normal esophageal tissue were detected by immunohistochemical S-P method. Results The expression of SKP2 protein in esophageal carcinoma was 64.4% (78/121), while it was 13.3% (12/90) in normal esophageal tissues. The difference was significant (P <0.05). The positive expression rate of SKP2 protein had no correlation with age, sex, tumor size, lymph node metastasis and TNM stage (P> 0.05), but correlated with tumor differentiation (P <0.05). There was a significant negative correlation between SKP2 and P27 expression (P <0.05). Conclusion It is demonstrated from the protein level that the abnormal expression of SKP2 promotes the development of esophageal cancer and may provide a new method for the early diagnosis and treatment of esophageal cancer.
