[目的]观察原白头翁素衍生物对人肝癌HepG-2细胞的诱导凋亡作用.[方法]应用MTT法、HE染色及AO/EB荧光染色方法观察原白头翁素衍生物对人肝癌HepG-2细胞形态的影响.[结果]原白头翁素衍生物对人肝癌HepG-2细胞增殖抑制作用呈质量浓度依赖性和时间依赖性(P<0.05,P<0.01).倒置显微镜下见,给予原白头翁素衍生物5.0 mg/L后早期细胞出现凋亡,晚期细胞发生碎裂坏死;HE染色和AO/EB荧光染色观察见细胞出现典型凋亡形态学变化.[结论]原白头翁素衍生物对人肝癌HepG-2细胞有抑制增殖和诱导凋亡作用.
[Objective] To observe the induction of apoptosis of hepatoma HepG-2 cells by original anemones derivative. [Methods] MTT assay, HE staining and AO / EB fluorescent staining were used to observe the effect of Pulsatillae on HepG- [Results] The original Pulsatilin derivatives inhibited the proliferation of human hepatocellular carcinoma HepG-2 cells in a concentration-dependent and time-dependent manner (P <0.05, P <0.01) .Inverted microscope, Apoptosis was observed in the early stage cells after 5.0 mg / L Su-derivatives and fragmentation and necrosis in the late stage cells. The morphological changes of typical cells were observed by HE staining and AO / EB fluorescent staining. [Conclusion] Liver cancer HepG-2 cells inhibit proliferation and induce apoptosis.
