目的建立一种三重荧光PCR检测手足口相关病毒的方法,并对收集的样本进行检测。方法分别用普通PCR和单荧光PCR进行引物探针的测试,然后将确定的引物和探针混合进行三重荧光PCR测试和标本的检测。结果建立了一种三重荧光PCR方法,利用该方法对94份标本进行检测,其中肠道病毒71型(EV71)37.23%,科萨奇病毒A16(COXA16)35.11%,其他肠道病毒4.26%。结论本研究建立的三重荧光PCR可以特异地检测出引起手足口病的EV71和COXA16以及其他肠道病毒。
Objective To establish a triplex fluorescence PCR method for detection of hand, foot and mouth-related viruses and to detect the collected samples. Methods Primer probes were tested by both ordinary PCR and single fluorescence PCR, respectively. Then, the specific primers and probes were mixed to carry out triplex fluorescence PCR test and specimen detection. Results A triplex fluorescent PCR method was established. 94 samples were detected by this method, including 37.23% of enterovirus 71 (EV71), 35.11% of Coxsackie virus A16 (COXA16) and 4.26% of other enteroviruses. Conclusion The triplex fluorescent PCR established in this study can specifically detect EV71 and COXA16, which cause hand-foot-and-mouth disease, as well as other enteroviruses.
