[目的]了解2010年德阳市手足口病患者中肠道病毒的主要血清型及EV71毒株的基因型别,为德阳市手足口病的预防控制提供科学依据。[方法]采用实时荧光PCR方法对四川省德阳市手足口病疑似病例的68份咽拭子或疱疹液标本进行肠道病毒通用型(EV)、肠道病毒71型(EV71)型和柯萨奇病毒A组16型(CoxA16)的病毒核酸检测。并对2份EV71阳性标本进行VP1区核苷酸序列测序和进化树分析。[结果]68份标本中,CoxA16阳性率41.18%(28/68),EV71阳性率17.65%(12/68),其他肠道病毒阳性率为7.35%(5/68)。2份EV71毒株VP1区的核苷酸序列进化树分析,证实均属C4基因亚型。[结论]2010年德阳市手足口病以CoxA16感染为主,EV71及其他非CoxA16、非EV71肠道病毒并存。2份德阳EV71毒株经VP1区测序证实为C4基因亚型,与中国内地流行株基因亚型一致。
[Objective] To understand the major serotypes of enterovirus and the genotypes of EV71 strains in Deyang City patients with hand-foot-mouth disease in 2010, and provide a scientific basis for the prevention and control of hand-foot-mouth disease in Deyang City. [Method] 68 samples of throat swab or herpes fluid from suspected cases of hand, foot and mouth disease in Deyang city of Sichuan province were tested by real-time fluorescence PCR for EV71, EV71, Virus Nucleic Acid Detection of Oddavirus Group A, Type 16 (CoxA16). Two VP71-positive samples were sequenced and phylogenetic tree was constructed. [Results] The positive rate of CoxA16 was 41.18% (28/68) in 68 samples, while the positive rate of EV71 was 17.65% (12/68). The positive rate of other enterovirus was 7.35% (5/68). Two EV71 strains VP1 region of the nucleotide sequence analysis of the evolutionary tree, confirmed to belong to the C4 gene subtypes. [Conclusion] In 2010, CoxA16 infection was the major hand-foot-mouth disease in Deyang City, and EV71 and other non-CoxA16 and non-EV71 enterovirus co-existed. Two copies of Deyang EV71 strain were confirmed to be C4 subtype by VP1 region sequencing, which is consistent with the gene subtype of the epidemic strain in Mainland China.
