用聚丙烯酰胺包埋,再以戊二醛交联联合固定了α淀粉酶、糖化酶和葡萄糖异构酶,对联合固定化的最适条件和联合固定化酶的性质进行了初步研究.结果表明联合固定化酶较固定化单酶更能发挥协同效应,能够将底物一步水解并转化为产物.文中所用的固定化方法与其他方法相比,固定化酶的稳定性、半衰期及酶固定化后活力均较高.在60℃、pH55时,α淀粉酶和糖化酶二酶的联合固定化酶的活力最高,其操作半衰期达到79h;在60℃、pH75时,α淀粉酶、糖化酶和葡萄糖异构酶三酶的联合固定化酶的活力最高,其操作半衰期达到90h.
After being encapsulated with polyacrylamide, the α-amylase, glucoamylase and glucose isomerase were immobilized by cross-linking of glutaraldehyde. The optimal conditions for co-immobilization and the properties of co-immobilized enzyme were studied. The results showed that the combination of immobilized enzyme and immobilized single enzyme more synergistic effect, the substrate can be one-step hydrolysis and conversion to the product. Immobilized method used in this article compared with other methods, the stability of the immobilized enzyme, half-life and high activity after enzyme immobilization. At 60 ℃, pH55, α-amylase and saccharifying enzyme combined immobilized enzyme had the highest activity and its half-life reached 79h. At 60 ℃ and pH75, the activity of α-amylase, glucoamylase and glucose The activity of enzyme-linked immobilized enzyme was the highest, and its half-life was 90h.
