目的探讨血管紧张素(Ang)(1~7)对氧化低密度脂蛋白(ox-LDL)诱导人血管内皮细胞单核细胞趋化蛋白1(MCP-1)表达的影响。方法原代培养人脐静脉内皮细胞(HUVEC),分别以不同终浓度的 Ang(1~7)(10、100、1000 nmol/L)和 ox-LDL[25、50、100 mg/L(蛋白质含量)]进行单独或联合刺激,并以 A-779[Ang(1~7)特异性受体阻断剂,终浓度为100nmol/L)]预处理,孵育24 h,以 ELISA 方法检测培养基上清液中 MCP-1蛋白含量,RT-PCR 法检测其 mRNA 的表达。结果 ox-LDL 使 MCP-1蛋白、基因表达增加,呈剂量依赖性(P<0.05~0.01);基础状态下,Ang(1~7)对 MCP-1表达无显著影响,但对 ox-LDL 诱导 MCP-1的表达增强有抑制作用(P<0.05~0.01);以 A-779预处理后,Ang(1~7)的作用消失。结论 Ang(1~7)对氧化低密度脂蛋白诱导的内皮细胞单核细胞趋化因子1升高具有显著的抑制作用,此作用是通过特异性受体介导的。
Objective To investigate the effect of angiotensin (1-7) on the expression of monocyte chemoattractant protein-1 (MCP-1) in human vascular endothelial cells induced by ox-LDL. Methods Primary human umbilical vein endothelial cells (HUVECs) were cultured in vitro. The effects of different concentrations of Ang (1-7) (10, 100, 1000 nmol / L) and ox-LDL [25, 50, 100 mg / (1: 7) specific receptor blockers at a final concentration of 100 nmol / L) for 24 h. ELISA was used to detect the growth of medium The content of MCP-1 protein in supernatant was detected by RT-PCR. Results Ox-LDL increased MCP-1 protein and gene expression in a dose-dependent manner (P <0.05-0.01). Under the basal conditions, Ang (1-7) had no significant effect on MCP- (P <0.05 ~ 0.01). After pretreatment with A-779, the effect of Ang (1-7) disappeared. Conclusion Ang (1-7) has a significant inhibitory effect on the increase of monocyte chemotactic factor 1 induced by oxidized low density lipoprotein, which is mediated through specific receptors.
