我院应用免疫组织化学方法,研究大肠癌中nm23-H_1、p~(53)的表达,并探讨它们的表达与大肠癌浸润程度、淋巴结转移状况以及它们之间的相互关系。128例中女性71例,男性57例,年龄32~81岁,病变部位:升结肠癌36例,横结肠癌6例,降结肠癌18例,乙状结肠癌21例,直肠癌47例。肿瘤最大直径1~11cm,平均5.3cm。大体分型:隆起型53例,溃疡型56例,浸润型19例。镜下组织学分类:管状腺痛103例,粘液腺癌16例,乳头状腺癌9例,Dukes分期;A期48例,B期24例,C期36例,D期20例。区域淋巴结转移者56例,无转移者72例。方法:全部切片经HE染色后行SP免疫组织化学染色DAB显色。全部数据采用X~2及相关性检验.结果:①nm23-H,的表达主要位于细胞浆内,胞核可见弱阳性。128例大肠癌nm23-H_1阳性表达94例,大肠癌淋巴结转移阳性nm23-H_1表达显著高于淋巴结转移阴性者(P<0.005),②p~(53)的表达定位于细胞核内,128例大肠癌中p~(53)阳性表达68例,本组大肠癌中p~(53)的表达肿瘤浸润浆膜外者高于浸润至肌层者(P<0.005),淋巴结转移阳性者p~(53)表达显著高于淋巴结转移阴性者,(P<0.01),③本组nm23-H_1阴性大肠癌中的p~(53)阳性率显著高于nm23-H_1阳性者(P<0.001),P~(53)的过表达与nm23-H_1低表达有关。
Our hospital applied immunohistochemistry to study the expression of nm23-H_1 and p53 in colorectal cancer, and to explore the relationship between their expression and colorectal cancer invasion, lymph node metastasis and their relationship. Of the 128 patients, 71 were males and 57 were males, aged 32 to 81 years. Lesions were: 36 cases of ascending colon cancer, 6 cases of horizontal colon cancer, 18 cases of descending colon cancer, 21 cases of sigmoid colon cancer, and 47 cases of rectal cancer. The maximum diameter of the tumor is 1~11cm with an average of 5.3cm. General classification: uplift 53 cases, ulcer type 56 cases, infiltration type 19 cases. Microscopic histological classification: 103 cases of tubular gland pain, 16 cases of mucinous adenocarcinoma, 9 cases of papillary adenocarcinoma, Dukes stage; 48 cases of A stage, 24 cases of B stage, 36 cases of C stage, 20 cases of D stage. There were 56 patients with regional lymph node metastases and 72 patients without metastases. METHODS: All sections were stained with HE and stained with SP immunohistochemistry DAB. All data using X ~ 2 and correlation test. RESULTS: The expression of 1nm23-H was mainly located in the cytoplasm and the nucleus was weakly positive. Among the 128 cases of colorectal cancer, the positive expression of nm23-H1 was 94 cases. The positive expression of nm23-H1 in lymph node metastasis of colorectal cancer was significantly higher than that in negative lymph node metastasis (P<0.005). The expression of 2p~(53) was located in the nucleus and 128 cases of colorectal cancer. The positive expression of p53 was found in 68 cases. In this group, the expression of p53 in colorectal cancer was higher than those infiltrated into the muscular layer (P<0.005), and the positive rate of lymph node metastasis was p53. Expression was significantly higher in patients with negative lymph node metastasis (P<0.01). The positive rate of p53 in nm23-H_1 negative colorectal cancer was significantly higher in the 3 groups than in those with positive nm23-H_1 (P<0.001). Overexpression of (53) is associated with low expression of nm23-H_1.
