目的:从红细胞膜组分的变化探讨补阳还五汤改善老龄大鼠血液黏度的作用机制。方法:40只Wistar大鼠,随机分为轻龄对照组、模型组、补阳还五汤高、低剂量组,每组10只。给药19 d后,取血测定血液流变学指标以及红细胞膜组分相关指标。结果:老龄大鼠血液黏度明显高于轻龄大鼠,而补阳还五汤可明显降低老龄大鼠的全血黏度和血浆黏度,同时可明显降低红细胞膜上胆固醇和丙二醛(MDA)含量,提高红细胞变形能力和红细胞膜上唾液酸含量、超氧化物歧化酶(SOD)活性。结论:补阳还五汤降低血液黏度的机制可能与影响红细胞膜上脂质代谢,提高抗氧化能力,减少氧自由基生成,以及改善红细胞膜上的负电荷荷电量等方面有关。
Objective: To investigate the mechanism of Buyang Huanwu decoction on improving blood viscosity of aging rats from the changes of erythrocyte membrane components. Methods: Forty Wistar rats were randomly divided into mild control group, model group, Buyang Huanwu Decoction high and low dose group, 10 rats in each group. After administration for 19 days, blood samples were collected to determine the indexes of hemorheology and erythrocyte membrane components. Results: The blood viscosity of aged rats was significantly higher than that of light-aged rats, while Buyang Huanwu Decoction could significantly reduce the whole blood viscosity and plasma viscosity of aging rats, and at the same time significantly lower the levels of cholesterol and malondialdehyde (MDA) Content, increase erythrocyte deformability and sialic acid content, erythrocyte membrane superoxide dismutase (SOD) activity. Conclusion: The mechanism of Buyang Huanwu Decoction in reducing blood viscosity may be related to influencing lipid metabolism on erythrocyte membrane, increasing antioxidant capacity, reducing the generation of oxygen free radicals and improving the negative charge charge on erythrocyte membrane.
