目的:建立同时测定大鼠血浆中油酸和亚油酸含量的气相色谱法。方法:采用毛细管气相色谱法,色谱柱为OV-17毛细管柱(30m×0.32mm×0.50μm),柱温220℃,氢火焰离子化检测器(FID)检测。结果:该法能将油酸甲酯和亚油酸甲酯有效分离,分别在23.80μg.mL-1~2380μg.mL-1(r=0.9996)、24.51μg.mL-1~2451μg.mL-1(r=0.9997)线性关系良好,高、中、低3个浓度的样品提取回收率在75.83%~84.17%,方法回收率在95.85%~107.1%,日内和日间精密度RSD均≤4.7%。结论:本法简单、准确,可作为大鼠血浆中油酸和亚油酸成分的检测方法,为进一步研究鸦胆子油及其制剂的药代动力学奠定基础。
OBJECTIVE: To establish a gas chromatographic method for simultaneous determination of oleic acid and linoleic acid in rat plasma. Methods: Capillary gas chromatography was used. The column was OV-17 capillary column (30m × 0.32mm × 0.50μm). The column temperature was 220 ℃ and detected by hydrogen flame ionization detector (FID). Results: The method was able to separate methyl oleate and methyl linoleate effectively from 23.80μg.mL-1 to 2380μg.mL-1 (r = 0.9996), 24.51μg.mL-1 to 2451μg.mL- 1 (r = 0.9997). The recoveries of three samples with high, medium and low concentrations ranged from 75.83% to 84.17%, and the recoveries ranged from 95.85% to 107.1%. The intra-day and inter-day RSDs were all ≤4.7 %. Conclusion: This method is simple and accurate and can be used as a method for the determination of oleic acid and linoleic acid in rat plasma, which lays the foundation for further study on the pharmacokinetics of Brucea javanica oil and its preparation.
