目的:研究硫酸乙酰肝素(heparan sulfate,HS)的体外抗黑色素瘤作用。方法:从培养的人乳腺上皮癌MCF-7细胞提取HS链,将HS以不同终浓度作用于人A375黑色素瘤细胞,倒置显微镜下观察受试细胞的形态变化;MTT法检测细胞生长抑制率;双染法观察其对细胞凋亡的影响;活性检测法测定细胞中酶caspase-3的变化。结果:HS可抑制A375细胞的生长和增殖,诱导细胞凋亡增加,并促进了caspase-3的活化。结论:来源于MCF-7细胞的HS具有较强的体外抗黑色素瘤作用。
Objective: To study the anti-melanoma activity of heparan sulfate (HS) in vitro. METHODS: HS chains were extracted from cultured human breast epithelial cancer MCF-7 cells. HS was treated with different final concentrations on human A375 melanoma cells. Morphological changes of the tested cells were observed under an inverted microscope; MTT assay was used to determine the cell growth inhibition rate. Double staining was used to observe its effect on cell apoptosis; activity assay was used to determine the changes of caspase-3 in cells. Results: HS inhibited the growth and proliferation of A375 cells, induced apoptosis, and promoted the activation of caspase-3. Conclusion: HS derived from MCF-7 cells has strong anti-melanoma activity in vitro.
