为探讨胰腺癌新抗原 SC 6的有关理化性质,用 Protein A-Sepharose 4B 柱纯化的单克隆抗体(单抗)SC 6吸附到 CNBr-Sepharose 4B 特异亲合层析柱上,纯化出相应的 SC 6抗原。此抗原经 SDS-PAGE 和 Western blotting 测定显示一条带,分子量为670 000,性质属糖蛋白,其中含有17种氨基酸。此抗原与 CA 19-9等多种抗原不同,在胰腺癌组织中的阳性表达率为91.2%。上述结果表明,该抗原是一种与胰腺癌相关的新肿瘤标志物,对此癌的深入探讨提供了新的资料,且为mRNA 分子克隆的筛选及检测提供了靶抗原。
To investigate the physicochemical properties of pancreatic cancer new antigen, SC 6, monoclonal antibody (mAb) SC 6 purified with Protein A-Sepharose 4B column was adsorbed onto CNBr-Sepharose 4B-specific affinity chromatography column to purify the corresponding SC. 6 antigens. The antigen was determined by SDS-PAGE and Western blotting to show a band with a molecular weight of 670 000. The property is a glycoprotein and contains 17 amino acids. Unlike antigens such as CA 19-9, this antigen has a positive expression rate of 91.2% in pancreatic cancer tissues. The above results indicate that the antigen is a novel tumor marker associated with pancreatic cancer. It provides new information for further investigation of this cancer and provides target antigens for the screening and detection of mRNA molecular cloning.
