目的探讨子痫前期患者胎盘组织中缺氧诱导因子-1α(Hypoxia-inducible factors-1α,HIF-1α)、黏蛋白1(Mucin1,MUC1)和基质金属蛋白酶-9(Matrix Metall oproteinase-9,MMP-9)的表达及意义。方法采用免疫组化ABC法和Western Blot法检测30例正常晚期妊娠孕妇(对照组)和50例子痫前期患者(子痫前期组,其中轻度22例、重度28例)胎盘组织中HIF-1α、MUC1和MMP-9的蛋白定位和表达。结果 HIF-1α主要在胎盘绒毛合体滋养细胞表达;轻、重度子痫前期组胎盘组织中HIF-1α的表达均高于对照组(P<0.01);MUC1主要表达于胎盘合体滋养细胞,子痫前期重度组MUC1表达显著高于轻度组及对照组(P<0.01);MMP-9在胎盘合体滋养细胞表达,轻度、重度子痫前期组均低于对照组(P<0.01),差异有统计学意义。轻度与重度子痫前期组之间MMP-9的表达差异无统计学意义(P>0.05);重度子痫前期组HIF-1α的表达与MUC1呈正相关(P<0.01),而MUC1的表达与MMP-9呈负相关(P<0.01)。结论 HIF-1α、MUC1、MMP-9共同参与子痫前期的病理生理改变,并与病情严重症程度有一定关系。
Objective To investigate the expression of Hypoxia-inducible factors-1α (HIF-1α), Mucin1 (MUC1) and Matrix Metalloproteinase-9 (MMPs) in placenta of preeclampsia -9) expression and significance. Methods Immunohistochemical ABC method and Western Blot were used to detect the expression of HIF-1α in placenta of 30 normal pregnant women (control group) and 50 preeclampsia patients (mild preeclampsia group, 22 mild cases and 28 severe cases) , MUC1 and MMP-9 protein localization and expression. Results HIF-1α was mainly expressed in placental syncytiotrophoblasts. The expression of HIF-1α in placenta of mild and severe preeclampsia group was higher than that in control group (P <0.01). MUC1 mainly expressed in placental syncytiotrophoblast, eclampsia The expression of MUC1 in pre-severe group was significantly higher than that in mild group and control group (P <0.01). The expression of MMP-9 in placental syncytiotrophoblasts was lower than that in mild pre-eclampsia group (P <0.01) There is statistical significance. There was no significant difference in the expression of MMP-9 between mild and severe preeclampsia (P> 0.05). The expression of HIF-1α was positively correlated with MUC1 in severe preeclampsia (P <0.01), while the expression of MUC1 Negatively correlated with MMP-9 (P <0.01). Conclusion HIF-1α, MUC1 and MMP-9 are involved in the pathophysiological changes of preeclampsia and have a certain relationship with the severity of illness.
