制备丹酚酸组分磷脂复合物,以丹酚酸组分与大豆磷脂的复合率为指标,采用单因素试验和正交试验筛选其工艺参数,结果可得丹酚酸组分磷脂复合物制备的最佳条件为以四氢呋喃为反应溶剂,反应时间3 h,反应物质量浓度5 g·L-1,药物与磷脂的反应投料质量比1∶1.5,反应温度40℃。所制备的丹酚酸组分磷脂复合物在水和不同pH缓冲液中的油水分配系数得到较大提高,DSC,XRD,FTIR等物相分析结果表明丹酚酸组分以无定形状态存在于磷脂中。
The salvianolic acid component phospholipid complex was prepared. The compounding ratio of salvianolic acid component and soybean phospholipid was used as an index, and the process parameters were screened by single factor test and orthogonal test. The results showed that the salvianolic acid component phospholipid complex could be prepared The optimal reaction conditions were tetrahydrofuran as reaction solvent, reaction time 3 h, reaction mass concentration 5 g · L-1, reaction mass ratio of drug to phospholipid 1: 1.5 and reaction temperature 40 ℃. The oil-water partition coefficient of the prepared salvianolic acid component phospholipid complex in water and different pH buffers is greatly increased. The phase analysis results of DSC, XRD and FTIR indicate that the salvianolic acid component exists in an amorphous state in Phospholipid.
