目的对一株软珊瑚共附生真菌进行次级代谢产物及生物活性研究。方法采用活性追踪分离的方法,利用硅胶柱层析、Sephadex LH-20凝胶柱层析和半制备HPLC等方法,对采自广西涠洲岛的肉芝软珊瑚Sarcophytonsp.中分离获得的杂色曲霉Aspergillus versicolor(ZJ-2008015)的发酵产物进行分离和纯化,利用NMR、MS等波谱方法,并与文献对照,确定化合物的结构;利用卤虫致死活性模型和抗菌活性模型评价化合物的生物活性。结果从该菌发酵液的乙酸乙酯提取物中分离鉴定了6个单体化合物,分别为:(+)-sydonic acid(1),expansol G(2),(+)-sydowic acid(3),cyclo(L-Trp-L-Phe)(4),homodimeric WIN 64821(5)和ergosta-6α-hydroxy-4,22-dien-3-one(6)。结论抗菌活性测试结果表明,没药烷型倍半萜类化合物1~3对白色葡萄球菌Staphylococcus albus和金黄色葡萄球菌Staphylococcus aureus显示较强的抗菌活性。
Objective To study the secondary metabolites and biological activities of a soft coral epiphytic fungus. Methods Reactive tracing method was used to separate the variegated color of Sarcophyton sp. Collected from soft-shelled coral in Weizhou Island, Guangxi Province by silica gel column chromatography, Sephadex LH-20 gel column chromatography and semi-preparative HPLC. The fermentation products of Aspergillus versicolor (ZJ-2008015) were isolated and purified. The structures of the compounds were determined by spectral methods such as NMR and MS, and compared with the literature. The bioactivities of the compounds were evaluated by lethal activity model and antimicrobial activity model. Results Six monomer compounds were isolated and identified from the ethyl acetate extracts of the fermentation broth of the strain, which were (+) - sydonic acid (1), expansol G (2), (+) - sydowic acid , cyclo (L-Trp-L-Phe) (4), homodimeric WIN 64821 (5) and ergosta-6α-hydroxy-4,22-dien-3-one (6). Conclusion The results of antibacterial activity test showed that myrothecyl-type sesquiterpenoids 1 to 3 showed strong antibacterial activity against Staphylococcus albus and Staphylococcus aureus.
