目的观察6-羟多巴(6-hydroxydopamine,6-OHDA)损伤大鼠黑质后迷走神经背核(dorsal motor nucleus ofthe vagus,10N)中乙酰胆碱转移酶(choline acetyltransferase,ChAT)和酪氨酸羟化酶(tyrosine hydroxylase,TH)含量的变化。方法 16只SD大鼠根据随机数字表分为6-OHDA大鼠组和对照组。6-OHDA大鼠组双侧黑质(substantia nigra,SN)内注射6-OHDA,对照组双侧SN内注射等体积的生理盐水。6周后断头取脑,Nissl染色观察SN和10N中的Nissl小体,免疫荧光和免疫印迹检测SN内TH和10N内ChAT与TH的表达。结果与对照组比较,6-OHDA大鼠组SN内TH阳性细胞数量从(54±5)个减少至(13±2)个(P<0.05),蛋白表达水平从0.62±0.13降低至0.34±0.11(P<0.05),10N中ChAT阳性细胞数量从(37±3)个减少到(26±5)个(P<0.05),TH阳性细胞数量从(6±3)个增加到(13±2)个(P<0.05),背侧延髓内ChAT的蛋白表达水平从0.07±0.02下降至0.05±0.02(P<0.05),TH蛋白表达水平从0.08±0.05增高到0.13±0.04(P<0.05)。结论 6-OHDA大鼠10N中ChAT和TH表达变化可能与帕金森病患者的胃排空障碍存在一定的联系。
Objective To observe the effects of choline acetyltransferase (ChAT) and tyrosine hydroxylation in the dorsal motor nucleus of the vagus (10N) after 6-hydroxydopamine (6-OHDA) Changes of tyrosine hydroxylase (TH) content. Methods Sixteen SD rats were divided into 6-OHDA group and control group according to random number table. 6-OHDA rats were injected with 6-OHDA into substantia nigra (SN) group, and bilateral control rats were injected with equal volumes of normal saline. Six weeks later, the brain was decapitated. The Nissl bodies in SN and 10N were observed by Nissl staining. The expression of ChAT and TH in SN and SN in SN were detected by immunofluorescence and immunoblotting. Results Compared with the control group, the number of TH positive cells in the SN of 6-OHDA group decreased from 54 ± 5 to 13 ± 2 (P <0.05), and the protein expression level decreased from 0.62 ± 0.13 to 0.34 ± The number of TH positive cells decreased from (37 ± 3) to (26 ± 5) and the number of TH positive cells increased from (6 ± 3) to (13 ± (P <0.05). The protein expression of ChAT in dorsal medulla decreased from 0.07 ± 0.02 to 0.05 ± 0.02 (P <0.05), and the expression of TH protein increased from 0.08 ± 0.05 to 0.13 ± 0.04 (P <0.05) ). Conclusion The expression of ChAT and TH in 10N of 6-OHDA rats may be related to gastric emptying disorder in patients with Parkinson’s disease.
