目的:阐明奇正消痛贴膏快速透皮吸收的作用机理。方法:选用Wistar大鼠,于大鼠背部皮肤涂搽药剂,应用透射电镜观察该药对大鼠皮肤超微结构的影响。结果:用药后使皮肤角质层细胞间距明显增大,细胞结构变得疏松,外层细胞脱落。该药进入角质层细胞内,与细胞内基质相作用,引起基底角质层肿胀和水化程度增加。停药后3~5d,表皮角质层可基本恢复正常。结论:奇正消痛贴膏可明显降低皮肤对药物的屏障作用,并使基底角质层水化程度增加,从而增加了透皮吸收。
Objective: To elucidate the mechanism of rapid transdermal absorption of Qizhenxiaoxiao plaster. METHODS: Wistar rats were used to apply the drug to the back skin of rats. Transmission electron microscope was used to observe the effect of the drug on the ultrastructure of rat skin. RESULTS: After the administration of the drug, the distance between the stratum corneum cells of the skin was significantly increased, the cell structure became loose, and the outer layer cells fell off. The drug enters the stratum corneum cells and interacts with the intracellular matrix, causing swelling and hydration of the basal stratum corneum. After 3 to 5 days after drug withdrawal, the epidermis keratinocytes can return to normal. Conclusion: Qizhenxiaoxiao plaster can significantly reduce the barrier effect of skin on drugs and increase the degree of hydration of the basal stratum corneum, thus increasing transdermal absorption.
