目的:研究腺病毒介导的人白细胞介素-24(Ad-mda-7/IL-24)基因对喉癌细胞Hep-2增殖抑制效应及机制。方法:将携带有hIL-24基因的重组复制缺陷型腺病毒(Ad-hIL-24)感染喉癌细胞Hep-2,并以正常人脐静脉内皮细胞HUVEC为对照,采用RT-PCR方法检测Hep-2、HUVEC细胞中外源性hIL-24表达,以及Bcl-2和Bax的变化;采用四甲基偶氮唑蓝(MTT)比色法检测细胞的生长和凋亡情况。结果:腺病毒介导的hIL-24mRNA能在Hep-2细胞和HUVEC细胞中表达;在Hep-2细胞中,抗凋亡分子Bcl-2表达降低,而促凋亡分子Bax表达增强;在HUVEC细胞中Bcl-2表达没有变化,Bax表达有所增强。MTT法显示Ad-hIL-24能明显抑制喉癌细胞Hep-2的生长。在Hep-2细胞中,显微镜下可见明显的细胞凋亡。结论:Ad-hIL-24能抑制喉癌细胞Hep-2生长和诱导Hep-2细胞凋亡,但对正常细胞无毒性作用。
Objective: To study the inhibitory effect of adenovirus-mediated human interleukin-24 (Ad-mda-7 / IL-24) on Hep-2 cell proliferation and its mechanism. Methods: Hep-2 cells were infected with recombinant replication-defective adenovirus carrying hIL-24 gene (Ad-hIL-24). HUVECs were transfected with normal human umbilical vein endothelial cells. RT- -2, exogenous hIL-24 expression in HUVEC cells and the changes of Bcl-2 and Bax. The growth and apoptosis of cells were detected by MTT assay. Results: Adenovirus-mediated hIL-24 mRNA was expressed in Hep-2 cells and HUVECs. In Hep-2 cells, Bcl-2 expression was down-regulated and Bax expression was up-regulated. In HUVEC Bcl-2 expression did not change in cells, Bax expression was enhanced. MTT assay showed that Ad-hIL-24 significantly inhibited the growth of laryngeal carcinoma Hep-2 cells. In Hep-2 cells, obvious apoptosis was observed under microscope. Conclusion: Ad-hIL-24 can inhibit the growth of Hep-2 cells and induce the apoptosis of Hep-2 cells in laryngeal carcinoma cells, but it is non-toxic to normal cells.
