目的:建立用高效液相色谱法测定血浆中泮托拉唑浓度的方法,为进行泮托拉唑的体内药物代谢、生物利用度研究提供基础。方法:血浆用0.01mol·L-1磷酸氢二钠做pH调节剂,二氯甲烷做提取溶剂,硝西泮作内标。采用PlatinumC18(5u,250mm×4.6mm)柱,流动相:甲醇水醋酸盐缓冲液(40∶60∶5),流速1.0ml·min-1,检测波长286nm。结果:该法回收率平均在98.36%~104.27%,重现性高,RSD在4.693%~9.12%范围内(n=5),线性范围:20~4000μg·L-1,最低检测浓度15μg·L-1。结论:HPLC测定血浆中泮托拉唑的方法简便、准确、灵敏、专一,能满足血药浓度测定及药代动力学研究的需要。
OBJECTIVE: To establish a method for the determination of pantoprazole concentration in plasma by HPLC and to provide a basis for the study of the drug metabolism and bioavailability of pantoprazole in vivo. Methods: The plasma was treated with 0.01 mol·L-1 sodium phosphate dibasic, pH adjuster, methylene chloride as extraction solvent, and nitrazepine as internal standard. Platinum C18 (5u, 250mm × 4.6mm) column was used. The mobile phase was methanol-water-acetate buffer (40:60:5), the flow rate was 1.0ml · min-1 and the detection wavelength was 286nm. Results: The average recoveries of this method ranged from 98.36% to 104.27% with high reproducibility, RSD ranged from 4.693% to 9.12% (n = 5), linear range from 20 to 4000 μg · L -1, the lowest detection concentration of 15μg · L-1. Conclusion: The HPLC method for the determination of pantoprazole in plasma is simple, accurate, sensitive and specific. It can meet the need of determination of plasma concentration and pharmacokinetics.
