目的探讨卵巢上皮性肿瘤中S期激酶相关蛋白2(Skp2)和第10号染色体同源缺失性磷酸酶-张力蛋白(PTEN)基因表达与其发生发展的关系。方法用免疫组织化学法检测卵巢良性肿瘤(30例)、卵巢交界性肿瘤(30例)及卵巢癌(50例)中Skp2、p27和PTEN蛋白的表达。结果 Skp2在卵巢癌中的表达(48.00%)明显高于良性肿瘤(0.00%)和交界性肿瘤(0.00%),差异有统计学意义(P<0.01);p27蛋白与PTEN蛋白在卵巢良性肿瘤(分别为90.00%、93.33%)和交界性肿瘤中(70.00%、70.00%)的表达明显高于卵巢癌(22.00%、40.00%),差异有统计学意义(P<0.01)。Skp2、p27和PTEN蛋白表达与卵巢癌的病理类型无关(P>0.05);卵巢癌中Skp2和p27的表达呈负相关(r=-0.637,P<0.01);Skp2和PTEN蛋白表达呈负相关(r=-0.726,P<0.01);p27和PTEN蛋白表达呈正相关(r=0.719,P<0.01)。结论 Skp2高表达、p27和PTEN蛋白低表达可能在卵巢上皮性肿瘤的发生发展中发挥重要作用,联合检测上述3种基因有助于判断卵巢癌的恶性程度和预后。
Objective To investigate the relationship between Skp2 and PTEN gene expression in epithelial ovarian tumors and its occurrence and development. Methods The expressions of Skp2, p27 and PTEN in benign ovarian tumors (30 cases), borderline ovarian tumors (30 cases) and ovarian cancer (50 cases) were detected by immunohistochemistry. Results The expression of Skp2 in ovarian cancer (48.00%) was significantly higher than that in benign tumors (0.00%) and borderline tumors (0.00%) (P <0.01). The positive rates of p27 protein and PTEN in benign ovarian tumors (90.00%, 93.33%, respectively) and borderline tumors (70.00%, 70.00%) were significantly higher than those in ovarian cancer (22.00%, 40.00%), the difference was statistically significant (P <0.01). The expression of Skp2, p27 and PTEN was not related to the pathological type of ovarian cancer (P> 0.05). The expression of Skp2 and p27 in ovarian cancer was negatively correlated (r = -0.637, P <0.01) (r = -0.726, P <0.01). There was a positive correlation between p27 and PTEN protein expression (r = 0.719, P <0.01). Conclusion High expression of Skp2, low expression of p27 and PTEN protein may play an important role in the development of ovarian epithelial neoplasms. Combined detection of these three genes may help to determine the malignancy and prognosis of ovarian cancer.
