目的:探讨石蜡包埋组织芯片技术的方法学及其在大肠癌研究中的意义.方法:选取病理科档案材料中44例大肠癌、13例癌旁结肠黏膜和26份大鼠结肠石蜡包埋组织,采用组织芯片仪及手工法制作208点列阵组织芯片,并进行he染色和mib-1抗体免疫组织化学染色.光镜观察并计数mib-1阳性细胞在各组的分布.结果:经染色后组织芯片大部结构完整,每一点阵均有代表性组织形态;mib-1阳性细胞标记指数(li)在中低分化大肠腺癌(50.17%)中明显高于高分化(管状)大肠腺癌(32.38%),而癌旁大肠黏膜li为10.08%.三者之间均具有非常显著性差异(p<0.01).结论:手工法组织芯片制作技术是一种简易可行的方法,在肿瘤病理研究中必将起到重要作用.mib-1阳性细胞标记指数与肿瘤细胞生长活跃程度相关,可作为判断大肠癌生物学行为及其预后的指标之一.
Objective: To explore the methodology of paraffin-embedded tissue microarray and its significance in the study of colorectal cancer.Methods: Forty-four patients with colorectal cancer, 13 patients with adjacent colonic mucosa and 26 rats with paraffin-embedded paraffin embedded in the pathological archives were enrolled. Tissue microarray and manual method were used to make 208 array microarrays, and he staining and mib-1 antibody immunohistochemical staining were performed.Mil-microscope observation and counting the distribution of mib-1 positive cells in each group.Results: After staining, most of the tissue microarrays were structurally intact and each dot matrix had representative histological morphology. The mib-1 positive cell labeling index (li) was significantly higher in moderately poorly differentiated colorectal adenocarcinomas (50.17%) than in highly differentiated (tubular) Adenocarcinoma (32.38%), while the adjacent colorectal mucosa li was 10.08% .There was a significant difference between the three (p <0.01) .Conclusion: Manual method tissue chip production technology is a simple and feasible method, Tumor pathology will play an important role.Mib-1 positive cell labeling index and the degree of tumor cell growth activity may be used as a judgment of biological behavior of colorectal cancer and prognosis of one of the indicators.
