目的:探讨糖肾颗粒对糖尿病大鼠肾组织核转录因子-κB(NF-κB)、单核细胞趋化因子1(MCP-1)表达的影响。方法:复制早期糖尿病肾病(DN)大鼠模型,将成模大鼠分为模型组,糖肾颗粒低、中、高剂量组和缬沙坦组,同时设正常对照组。观察大鼠一般状况、生化指标及肾组织病理,通过实时荧光定量PCR和Westernblot技术检测NF-κB和MCP-1 mRNA和蛋白表达,ELISA技术检测血中血栓素A2(TXA2)和前列环素(PGI2)含量。结果:模型组大鼠空腹血糖(FBG)升高,甘油三酯(TG)和胆固醇(TC)增加,尿微量白蛋白排泄率(U-alb)增加,内生肌酐清除率(Ccr)降低(P<0.01,P<0.05);TXA2、NF-κB、MCP-1明显升高,PGI2降低(P<0.05,P<0.01);经糖肾颗粒及缬沙坦干预后各项指标均有不同程度的好转。结论:糖肾颗粒通过降低NF-кB、MCP-1的表达抑制炎性反应从而对糖尿病肾脏损伤有保护作用。
Objective: To investigate the effect of Tangshen Granule on the expression of nuclear factor-κB and monocyte chemotactic factor-1 (MCP-1) in kidney of diabetic rats. Methods: The rat models of early diabetic nephropathy (DN) were duplicated. The model rats were divided into model group, Tangshen Granules in low, medium and high dose groups and valsartan group. At the same time, normal control group was established. The general condition, biochemical indexes and renal histopathology of rats were observed. The mRNA and protein expressions of NF-κB and MCP-1 were detected by real-time fluorescence quantitative PCR and Western blot. The levels of TXA2 and prostacyclin PGI2) content. Results: The fasting blood glucose (FBG), triglyceride (TG) and cholesterol (TC), urinary albumin excretion (U-alb) and endogenous creatinine clearance (Ccr) P <0.01, P <0.05). The levels of TXA2, NF-κB and MCP-1 were significantly increased and PGI2 was decreased (P <0.05, P <0.01) The degree of improvement. Conclusion: Tangshen Granule can protect diabetic nephropathy by reducing the expression of NF-κB and MCP-1 and inhibiting the inflammatory reaction.
