目的:为了研究体外培养中交感神经纤维或神经终末与心肌细胞之间的关系。方法:将新生大鼠的交感神经节与心肌细胞进行联合培养,用相差显微镜观察神经元的生长,用扫描电子显微镜观察神经肌肉连接的形成。结果:交感神经节与心肌细胞联合培养72~96小时,在相差显微镜下可观察到神经突起终止于搏动的心肌细胞表面。在扫描电子显微镜下可观察到神经纤维粗细不一,在心肌细胞表面交织成网状。有的神经纤维末端膨大并贴于心肌细胞表面。结论:这一研究建立了在体外培养中具有选择性交感神经支配的心肌细胞模型。
AIM: To investigate the relationship between sympathetic nerve fibers or neural terminals and cardiomyocytes in vitro. Methods: The sympathetic ganglia of neonatal rats were cultured with cardiomyocytes. The growth of neurons was observed by phase contrast microscopy. The formation of neuromuscular junctions was observed by scanning electron microscopy. RESULTS: Sympathetic ganglia were cultured with cardiomyocytes for 72-96 hours. The neurites were observed to terminate at the surface of pulsatile cardiomyocytes under a phase-contrast microscope. In the scanning electron microscope can be observed nerve fibers vary in thickness, in the myocardial cell surface interwoven into a mesh. Some ends of nerve fibers are swollen and attached to the surface of cardiomyocytes. Conclusion: This study established a model of cardiomyocytes with selective sympathetic innervation in vitro.
