目的探讨干细胞标记物LGR5(富含亮氨酸重复单位的G蛋白偶联受体5)蛋白在人结直肠癌组织中的表达水平及其与临床病理特征、结直肠癌演进过程的关系和意义。方法采用免疫组化法分别检测10例结直肠正常组织、59例腺瘤组织、70例结直肠癌标本中LGR5的表达,并分析其与临床病理指标的关系。结果①LGR5在结直肠肿瘤中表达水平明显高于结直肠正常组织,亦明显高于结直肠腺瘤(P<0.05);②结直肠癌组织中LGR5的表达水平与Dukes分期、淋巴结转移呈正相关,与组织分型、分化程度、临床特征无相关性;③结直肠腺瘤中LGR5的表达水平与临床病理参数均无相关性。结论LGR5的表达说明结直肠癌的形成与干细胞相关,LGR5在肿瘤发生过程中可能起一定作用,且与肿瘤的进展相关,可以作为早期诊断的分子标记物。
Objective To investigate the expression of stem cell marker LGR5 (leucine-rich repeat-associated G protein-coupled receptor 5) protein in human colorectal cancer and its relationship with clinicopathological characteristics and the evolution of colorectal cancer . Methods The expression of LGR5 in 10 cases of normal colorectal tissues, 59 cases of adenomas and 70 cases of colorectal carcinomas was detected by immunohistochemistry. The relationship between the expression of LGR5 and clinicopathological parameters was analyzed. Results ① The expression of LGR5 in colorectal neoplasms was significantly higher than that in colorectal tissues and also significantly higher than that in colorectal adenomas (P <0.05). ② The expression of LGR5 in colorectal cancer was positively correlated with Dukes stage and lymph node metastasis, There was no correlation between the expression of LGR5 and clinicopathological parameters in colorectal adenoma. Conclusion The expression of LGR5 suggests that the formation of colorectal cancer is related to stem cells. LGR5 may play a role in tumorigenesis and may be associated with tumor progression and may serve as a molecular marker for early diagnosis.
