目的评价外源性PTEN抑癌基因对人A549肺癌细胞辐射敏感性的影响。方法体外培养A549肺癌细胞株,用脂质体介导法将PTEN导入A549肺癌细胞系,以空载体转染细胞作为对照。然后用MTT法和克隆形成实验测定2种癌细胞对~(60)Co-γ射线的敏感性。并检测肿瘤细胞PKB的表达。结果相同剂量照射下,A549-pBP-PTEN细胞存活与A549细胞相比明显减少,治疗指数分别为1.69;转染PTEN对肿瘤细胞PKB的表达产生抑制。结论外源性野生型PTEN抑癌基因能增强A549肺癌细胞对γ射线的敏感性。
Objective To evaluate the effect of exogenous PTEN gene on the radiosensitivity of human lung adenocarcinoma A549 cells. Methods A549 lung cancer cell lines were cultured in vitro. PTEN was transfected into A549 lung cancer cell lines by lipofectamine and transfected with empty vector as control. The sensitivity of the two kinds of cancer cells to ~ (60) Co-γ-rays was then determined by MTT assay and colony formation assay. The expression of PKB in tumor cells was detected. Results The survival rate of A549-pBP-PTEN cells was significantly decreased compared with that of A549 cells at the same dose of irradiation, the therapeutic index was 1.69, respectively. The transfection of PTEN inhibited the expression of PKB in tumor cells. Conclusion Exogenous wild-type PTEN tumor suppressor gene can enhance the sensitivity of A549 lung cancer cells to γ-rays.
