目的:探讨高血压大鼠心肌组织中胰岛素样生长因子-1(IGF-1)和缝隙连接蛋白43(CX43)的表达,并运用福辛普利进行干预,研究其心肌保护作用。方法:32只健康雄性SD大鼠随机分为空白对照组(n=8)、假手术组(n=8)以及模型组(n=16),模型组以二肾一夹的方法建立,模型建立成功后,随机分为福辛普利治疗组(n=8)和模型对照组(n=8)。治疗组给予福辛普利按照10mg/(kg·d)剂量灌胃,空白对照组、假手术组以及模型对照组给予相同体积的生理盐水灌胃,每日1次。给药8周后,麻醉处死动物,称体重(BW)、心脏(HW)和左心室(含室间隔LVW)重量,计算HW/BW为心脏指数(mg/g),LVW/BW为左心室指数(mg/g),SP免疫组化法检测心室肌组织中IGF-1、缝隙连接蛋白43(CX43)的表达。结果:与空白对照组相比,假手术组血压、心脏指数、左心室指数、IGF-1蛋白表达率、CX43含量变化不明显,模型组则明显升高(P<0.05);与模型对照组相比,福辛普利治疗组上述指标则改善明显(P<0.05)。结论:肾血管性高血压大鼠心室肌组织IGF-1蛋白表达增多,CX43蛋白表达增多,心脏指数及左心室指数增大,运用血管紧张素转换酶抑制剂福辛普利干预后,IGF-1蛋白表达率、CX43蛋白表达减少,心功能有所改善,提示福辛普利可能通过抑制IGF-1表达、降低CX43表达来改善心功能。
AIM: To investigate the expression of insulin-like growth factor-1 (IGF-1) and connexin43 (CX43) in myocardium of hypertensive rats and to study the protective effect of Fosinopril on myocardial protection. Methods: Thirty-two healthy male Sprague-Dawley rats were randomly divided into blank control group (n = 8), sham operation group (n = 8) and model group (n = 16) After successful establishment, they were randomly divided into fosinopril treatment group (n = 8) and model control group (n = 8). The patients in the treatment group were given fosinopril orally at a dose of 10 mg / (kg · d), while the blank control group, sham operation group and model control group were given the same volume of physiological saline once a day. Eight weeks after the administration, the animals were anesthetized and weighed (BW), heart (HW), and left ventricle (including ventricular septum LVW) weights. HW / BW was calculated as cardiac index Index (mg / g) and SP immunohistochemistry were used to detect the expression of IGF-1 and connexin43 (CX43) in ventricular myocytes. Results: Compared with the blank control group, the blood pressure, cardiac index, left ventricular index, IGF-1 protein expression and CX43 content in the sham operation group were not significantly changed, but the model group was significantly increased (P <0.05) Compared with fosinopril treatment group, the above indexes improved significantly (P <0.05). Conclusions: The expression of IGF-1 protein in myocardium of renovascular hypertensive rats increased, the expression of CX43 protein increased, and the cardiac index and left ventricular index increased. After intervention with fosinopril, an angiotensin converting enzyme inhibitor, IGF- 1 protein expression, CX43 protein expression decreased cardiac function improved, suggesting that fosinopril may inhibit IGF-1 expression, reduce CX43 expression to improve cardiac function.
