目的检测某铀矿氡职业暴露人群痰细胞中6-氧-甲基嘌呤-DNA甲基转移酶(MGMT)基因和p16基因的甲基化状态,为寻找氡致肺癌高危人群的分子标记物提供试验依据。方法91名氡职业暴露工人按氡子体累积暴露剂量工作水平月(WLM)分为高(>120 WLM)、中(60~120 WLM)、低(30~60 WLM)和较低(2~30 WLM)4个剂量组,用聚合酶链反应-甲基化特异性(MSP)检测4组人群痰细胞中的p16和MGMT基因的异常甲基化状态。结果随着氡子体累积暴露剂量的增加,p16基因甲基化率(0.00%~20.00%)、MGMT基因甲基化率(0.00%~28.00%)、总甲基化率(0.00%~40.00%)均呈明显上升趋势,差异有统计学意义(P<0.01)。结论p16和MGMT基因甲基化与氡子体累积暴露剂量有关。
Objective To detect the methylation status of 6-O-methyl-purine-DNA methyltransferase (MGMT) gene and p16 gene in sputum cells of workers exposed to radon in a uranium mine, and to provide molecular markers for looking for high risk population of lung cancer caused by radon Test basis. Methods Ninety-one radon occupational exposure workers were divided into high (> 120 WLM), moderate (60-120 WLM), low (30-60 WLM) and low (2-60 WLM) 30 WLM) were used to detect the abnormal methylation status of p16 and MGMT gene in sputum cells of 4 groups by polymerase chain reaction-methylation specificity (MSP). Results The results showed that the rates of methylation of p16 gene (0.00% -20.00%), methylation of MGMT gene (0.00% -28.00%) and total methylation rate (0.00% -40.00) %) Showed a significant upward trend, the difference was statistically significant (P <0.01). Conclusion The methylation of p16 and MGMT genes is related to the cumulative exposure dose of radon.
