探索了用HPLC法分离、测定6种熊胆样品中3种主要磷脂组合,即磷脂酰胆碱(PC)、磷脂酰肌醇(PI)、磷脂酰乙醇胺(PE)的方法。在选定的色谱条件下,组分PC的平均回收率为88.50%。重现性实验中,PC保留时间的相对标准差(RSD)为0.79%,峰面积的RSD为7.24%。同时用正交实验对从熊胆样品中提取磷脂的方法进行了优化,得到最佳提取条件是:提取液氯仿和甲醇的配比为1∶1,提取液与样品体积比为25∶1,混合后涡旋4min,用于洗涤的饱和NaCl溶液与样品液体积比为1∶2。
The method for the separation and determination of the three major phospholipid combinations in six kinds of bear gall samples by HPLC, namely phosphatidylcholine (PC), phosphatidylinositol (PI) and phosphatidylethanolamine (PE), was explored. Under the selected chromatographic conditions, the average recoveries of component PC were 88.50%. In reproducible experiments, the relative standard deviation (RSD) of PC retention time was 0.79%, and the RSD of peak area was 7.24%. At the same time, the orthogonal experiment was used to optimize the method of extracting phospholipids from bear gall samples. The optimal extraction conditions were as follows: the ratio of chloroform to methanol was 1: 1, the volume ratio of extract to sample was 25: 1, After mixing vortex 4min, saturated NaCl solution for washing and sample volume ratio of 1: 2.
