目的:探讨部分长链非编码RNA(lncRNA)在中耳胆脂瘤上皮组织与正常外耳道皮肤组织中是否存在表达差异。方法:采用实时荧光定量PCR方法,分别检测25例中耳胆脂瘤上皮组织及15例正常外耳道皮肤组织样本中6种lncRNA(HOTAIR、ANCR、TINCR、PRINS、BANCR、PICSAR)的表达情况;并对比不同中耳胆脂瘤骨质破坏程度的患者之间,其lncRNA的表达水平是否存在差异。结果:相对于正常外耳道皮肤组织,中耳胆脂瘤上皮组织中HOTAIR表达水平显著升高,且差异有统计学意义(P<0.01),其余5种lncRNA在中耳胆脂瘤与正常外耳道皮肤组织中的表达差异无统计学意义(P>0.05);而不同骨质破坏程度的胆脂瘤患者之间,其肿瘤组织HOTAIR表达水平差异无统计学意义(P>0.05)。结论:中耳胆脂瘤上皮组织中HOTAIR的表达较正常外耳道皮肤组织上调,其上调水平与胆脂瘤对周围骨质破坏程度之间无明显相关。
Objective: To investigate the expression differences of some long-chain non-coding RNAs (lncRNAs) in middle ear cholesteatoma epithelial tissues and normal external auditory canal skin tissues. Methods: The expression of 6 kinds of lncRNA (HOTAIR, ANCR, TINCR, PRINS, BANCR, PICSAR) in 25 cases of middle ear cholesteatoma epithelial tissue and 15 cases of normal external auditory canal skin tissue were detected by real-time fluorescence quantitative PCR. There was a difference in the expression levels of lncRNA between patients with different types of middle ear cholesteatoma. Results: Compared with normal external auditory canal skin tissue, the expression of HOTAIR in middle ear cholesteatoma epithelium was significantly increased (P <0.01), and the other 5 kinds of lncRNA in middle ear cholesteatoma and normal external auditory canal skin (P> 0.05). However, there was no significant difference in HOTAIR expression between the patients with different degrees of bone destruction (P> 0.05). CONCLUSION: The expression of HOTAIR in the middle ear cholesteatoma epithelium is up-regulated compared with that of the normal external auditory canal. The up-regulation of HOTAIR has no significant correlation with the degree of bone destruction.
