目的探讨血管内皮生长因子(Vascular endothelial growth factor,VEGFmRNA)对糖尿病股骨头不同组织的旁分泌作用。方法建立速发型链脲佐菌素(streptozotocin,STZ)糖尿病大鼠模型,随机分为正常5周组(CON1)、10周组(CON2)及15周组(CON3),糖尿病5周组(DM1)、10周组(DM2)及15周组(DM3),每组10只。对各组大鼠股骨头关节软骨、生长板、股骨头松质骨光镜下作组织病理学分析;观察分析血管内皮生长因子(VEGFmRNA)在不同组织原位杂交的表达强度。结果 VEGFmRNA在对照各组的关节软骨均有表达,糖尿病组随病程延长,关节软骨与松质骨VEGFmRNA表达均高于正常组(<0.01)。糖尿病15周组生长板VEGFmRNA表达均高于正常组(<0.01)。糖尿病各组股骨头松质骨VEGFmRNA表达逐渐增强(<0.05)。结论糖尿病大鼠股骨头关节软骨VEGFmRNA旁分泌功能最活跃,可能参与糖尿病发病。
Objective To investigate the paracrine effect of vascular endothelial growth factor (VEGF) on different tissues of diabetic femoral head. Methods A rat model of rapid onset streptozotocin (STZ) was established and randomly divided into 5 groups: CON1, CON2, CON3, DM1 ), 10 weeks group (DM2) and 15 weeks group (DM3), 10 rats in each group. Histopathological analysis of the femoral head articular cartilage, growth plate and femoral head cancellous bone in rats of each group was performed. The expression intensity of vascular endothelial growth factor (VEGF mRNA) in situ hybridization was observed and analyzed. Results VEGFmRNA was expressed in articular cartilage of all the control groups. The expression of VEGFmRNA in articular cartilage and cancellous bone in diabetic group was longer than that in normal group (P <0.01). The expression of VEGF mRNA in the growth plate of 15-week diabetes mellitus group was higher than that in the normal group (<0.01). The expression of VEGFmRNA in cancellous bone of diabetic rats increased gradually (P <0.05). Conclusion Diabetic rat femoral articular cartilage VEGFmRNA paracrine most active, may be involved in the pathogenesis of diabetes.
