目的:分析中药化学成分分析、分离技术及策略。方法:采用色谱质谱联用技术分析中药天葵子95%乙醇提取部位化学分成。通过柱层析技术对醇提部位化学成分进行分离处理,利用CAPCELL PAK C18(250mm×4.6mmI.D.,5μm)色谱柱、紫外图谱与质谱对单体结构进行检测、鉴定。结果:通过正离子质谱信息分析对天葵子醇提中的六个化合物进行鉴定。包括东方唐宋草甘、冬青氰苷、2-丙烯酸-3(4’-羟基苯基)-(4“-羧基苯基)、Ehretioside B(B对照品)、苦瓜酚苷A、脂。通过柱层析分离出三个单体,对cirsiumaldehyde(蓟醛)、小檗碱、唐松草酚定进行鉴定,首先从天葵子中分离出来的是小檗碱。结论:通过色谱质谱联用技术、现代柱色谱分离、谱波解析,对天葵子中化学成分进行定性分析、分离,这种方法具有一定的可行性与合理性,且结果准确,值得推广和临床应用。
Objective: To analyze the chemical composition of traditional Chinese medicine, separation techniques and strategies. Methods: Chromatography-mass spectrometry (GC-MS / MS) was used to analyze the chemical constituents of 95% ethanol extracted from Chinese sunflower seed. The chemical constituents of alcohol extraction were separated by column chromatography. The structure of the monomer was detected and identified by CAPCELL PAK C18 (250mm × 4.6mmI.D., 5μm) column, UV spectrum and mass spectrum. Results: Six compounds identified in the extract of spinosad were identified by positive ion mass spectral analysis. Including oriental Tang and Song glycyrrhizin, hollycinoside, 2-acrylic acid-3 (4’-hydroxyphenyl) - (4 ”- carboxyphenyl), Ehretioside B (B reference substance), bitter gourd glycosides A, fat. Three monomers were separated by column chromatography and identified as cirsium, catechol, berberine, and triclopidine, and berberine was first isolated from the sunflower seed.Conclusion: Chromatography-mass spectrometry Technology, modern column chromatography, spectral analysis, the chemical composition of the sunflower seed qualitative analysis, separation, this method has a certain feasibility and rationality, and the results are accurate, it is worth promoting and clinical application.
