目的研究粉尘螨(Der.f)变应原对树突状细胞的作用。方法分别检测经Der.f变应原作用后,DC2.4(树突状细胞株)细胞白介素-12(IL-12)和IL-10的分泌,TLR2受体的基因表达和丝裂原活化蛋白激酶(MAPK)p44/42的磷酸化水平。结果Der.f促进DC2.4IL-10的分泌(由空白组的不分泌到Der.f作用组的27.7pg/mL,P<0.01),抑制IL-12的分泌(由空白组的22.8pg/mL到Der.f作用组的15.9pg/mL,P<0.05),并可促进p44/42MAPK蛋白的磷酸化和TLR2的基因表达,U0126可逆转其细胞因子的分泌模式,并降低p44/42MAPK磷酸化水平。结论Der.f变应原可影响树突状细胞的细胞因子分泌,受体表达及信号通路磷酸化水平。
Objective To study the effect of Der.f allergen on dendritic cells.
