采用流式细胞术,视频观察和琼脂糖凝胶电泳,观察糖皮质激素曲安奈德预处理对紫杉醇等诱发的人乳癌细胞凋亡反应的影响.曲安奈德预处理可抑制紫杉醇诱导的细胞凋亡,流式细胞术和视频观察显示凋亡抑制率分别为49.3%和52.0%.琼脂糖凝胶电泳显示曲安奈德抑制抗微管剂紫杉醇,秋水仙碱,长春碱所引起的DNA裂解反应,“阶梯状”DNA条带密度降低.对DNA和蛋白质合成抑制剂则无明显影响.甾醇类激素预处理对紫杉醇无明显抑制作用.研究结果提示曲安奈德预处理对紫杉醇诱导的乳癌细胞凋亡具有阻抑作用
Flow cytometry, video observation and agarose gel electrophoresis were used to observe the effects of glucocorticoid triamcinolone acetonide on paclitaxel-induced apoptosis in human breast cancer cells. Triamcinolone acetonide pretreatment can inhibit paclitaxel-induced apoptosis, flow cytometry and video observation showed that apoptosis inhibition rates were 49.3% and 52.0%. Agarose gel electrophoresis showed that triamcinolone acetonide inhibited DNA cleavage reaction caused by antimicrotubules paclitaxel, colchicine and vinblastine, and the density of “ladder” DNA bands was decreased. No significant effect on DNA and protein synthesis inhibitors. Steroid hormone pretreatment did not significantly inhibit paclitaxel. The results suggest that triamcinolone acetonide pretreatment inhibited paclitaxel-induced apoptosis in breast cancer cells
