DNA甲基化是生物基因组最常见,也是最重要的表观遗传修饰形式之一,随着对DNA甲基化研究的不断深入,由AFLP技术发展而来,基于PCR检测的DNA甲基化敏感扩增多态性(MSAP)技术,凭借其应用广泛,操作简便等优点成为检测DNA甲基化水平的有力工具。目前已被广泛应用于种质资源鉴定、品种改良、基因表达调控等许多领域。本文对MSAP技术的原理、操作流程及其在玉米遗传育种研究中的各项研究进展进行了综述,并对该技术今后的应用前景进行了展望。
DNA methylation is one of the most common and important forms of epigenetic modification in the genome of the genomic DNA. With the deepening of the research on DNA methylation, DNA methylation has been developed from AFLP technology and based on PCR Amplified polymorphism (MSAP) technology, with its wide range of applications, easy to operate and other advantages as a powerful tool to detect DNA methylation levels. Has been widely used in germplasm identification, variety improvement, gene expression regulation and many other fields. In this paper, the principle of MSAP technology, its operation procedure and its research progress in maize genetics and breeding are reviewed, and the future application prospects of this technology are prospected.
