目的:观察胃溃疡肝郁脾虚证大鼠胃组织差异性基因表达的情况,验证病证结合大鼠存在此类特异性基因表达,明确存在低氧微环境,探讨柴黄胃溃宁干预各基因表达的疗效及机制。方法:以多因素复合复建胃溃疡肝郁脾虚证大鼠模型,用Real-time PCR技术检测柴黄胃溃宁干预模型大鼠差异性基因的表达。结果:模型组大鼠差异性基因M、P、F、I较正常组低表达(P<0.05)。中药干预后,柴黄胃溃宁各剂量组影响M、P、F、I基因出现不同程度的高表达,其中M、P基因中黄芪组作用最明显(P<0.05),I、F基因柴黄胃溃宁高剂量组作用最明显(P<0.05)。结论:病证结合模型确有呼吸链基因改变情况,证实胃溃疡肝郁脾虚状态发生中存在低氧的情况,验方柴黄胃溃宁对该环境中差异性基因表达有明显调节作用,可能通过改善低氧环境起效。
OBJECTIVE: To observe the differential expression of differentially expressed genes in gastric tissue of gastric ulcer rats with liver-qi deficiency and spleen-qi syndrome, and to verify the existence of such specific gene expression in rats with pathological syndromes, and to confirm the existence of hypoxic microenvironment. Efficacy and mechanism of expression. Methods: The rat model of gastric ulcer due to liver-qi and spleen-deficiency syndrome was reconstructed by multiple factors, and the expression of differential genes was detected by real-time PCR in the rat model of Chaihuang-wei-suing-ning. Results: The expression of M, P, F and I genes in model group was lower than that in normal group (P <0.05). CONCLUSIONS: The disease-syndrome-binding model indeed has the change of respiratory chain gene, confirming the existence of hypoxia in the state of gastric ulcer due to liver depression and spleen deficiency. The prescription Fang Chai Weicuning significantly regulates the differential gene expression in this environment, possibly through Improve hypoxia environment effect.
