目的:建立血浆中多西紫杉醇浓度的高效液相色谱测定方法。方法:以地西泮为内标,血浆样品经叔丁基甲醚3次提取,采用大连依利特C18(250mm×4.6mm,5μm)色谱分析柱;流动相为乙腈-水(47∶53);流速为1.0mL.min-1;检测波长为230nm;进样量为50μL。结果:地西泮和多西紫杉醇的保留时间分别是10.9min和13.2min,多西紫杉醇的最低检测质量浓度为20μg.L-1,血浆样品质量浓度在0.25~16.5mg.L-1范围内与峰面积比线性关系良好,回归方程为C=10.117X+0.428,r=0.9998(n=7),提取回收率为87.24%~92.64%(n=5),方法回收率为97.52%~101.44%,日内RSD为3.6%,2.4%,0.79%,日间RSD为3.8%,2.7%,1.4%,且稳定性良好。结论:本方法简便灵敏,干扰小,可用于多西紫杉醇血药浓度的测定。
Objective: To establish a method for the determination of docetaxel in plasma by high performance liquid chromatography. Methods: The plasma samples were extracted with tert-butyl methyl ether three times with diazepam as the internal standard. The HPLC analysis was carried out on a Dalian elite C18 column (250 mm × 4.6 mm, 5 μm). The mobile phase consisted of acetonitrile-water (47:53) 1.0mL.min-1; detection wavelength of 230nm; injection volume of 50μL. Results: The retention time of diazepam and docetaxel were 10.9min and 13.2min respectively, the lowest concentration of docetaxel was 20μg.L-1 and the concentration of plasma samples was within the range of 0.25-16.5mg.L-1 The regression equation was C = 10.117X + 0.428, r = 0.9998 (n = 7), the recoveries were 87.24% ~ 92.64% (n = 5) and the recoveries were 97.52% ~ 101.44 %, Intraday RSD was 3.6%, 2.4%, 0.79%, daytime RSD was 3.8%, 2.7%, 1.4%, and the stability is good. Conclusion: The method is simple and sensitive, the interference is small, can be used for the determination of docetaxel blood concentration.
