目的 研究肝细胞色素P4502E1在实验性肝纤维化组织中的表达。方法 8只Wistar大鼠,建立四氯化碳(CCl4)肝纤维化模型,用免疫组织化学方法观察肝纤维化模型肝组织中肝细胞色素P4502E1。结果 正常肝组织内,CYP2E1表达仅见于中央静脉周围区,主要表达于肝腺泡Ⅲ区,2~3层细胞厚,肝细胞浆和细胞膜可见CYP2E1表达。在CCl4模型肝组织中,CYP2E1表达的强度增加,分布的方式亦发生改变,可见于肝腺泡Ⅰ和Ⅱ区,与肝细胞脂肪变分布相一致,在假小叶的肝细胞中不见其表达。CYP2E1在肝细胞内分布以胞浆型为主,亦可见膜型和核型。8只实验动物肝纤维化组织中,2只强阳性,3只中等程度阳性,3只弱阳性。结论 CYP2E1表达与肝纤维化的发生有关,动物个体间存在CYP2E1多态性。
Objective To study the expression of hepatocyte cytochrome P450 2E1 in experimental liver fibrosis. Methods Eight Wistar rats were divided into four groups: CCl4-induced hepatic fibrosis model and hepatic cytochrome P4502E1 expression in hepatic fibrosis model were observed by immunohistochemical method. Results The expression of CYP2E1 in normal liver tissue was only found in the central venous region, which was mainly expressed in hepatic acinar area Ⅲ. In the CCl4 model liver tissue, the expression of CYP2E1 increased in intensity and the distribution pattern also changed. It was found in the hepatic acinus Ⅰ and Ⅱ area, consistent with the distribution of fatty degeneration in hepatocytes, but not in the hepatocytes of pseudolobules. Cytoplasmic distribution of CYP2E1 in hepatocytes mainly cytoplasm, also can be seen membranous and karyotype. Of the 8 experimental animals, 2 were strongly positive in liver fibrosis, 3 were moderately positive and 3 were weakly positive. Conclusion The expression of CYP2E1 is related to the occurrence of liver fibrosis. CYP2E1 polymorphism exists in animal individuals.
