目的 :探讨培养条件及冻存方法对肺腺癌细胞株的影响。方法 :在培养基中放弃使用抗生素 ,控制细胞传代、消化条件 ,采用快速冻存法保存细胞。结果 :37℃ ,5 % CO2 孵箱培养 2 h左右开始贴壁 ,8h左右完全贴壁 ,48~6 0 h可形成单层。结论 :(1)只要切实注意无菌操作规程 ,培养基中无需常规使用抗生素 ;(2 )控制胰酶条件、离心速度可减少细胞培养过程中的损伤 ;(3)细胞快速冻存法优于传统的慢速冻存法。
Objective: To explore the effects of culture conditions and cryopreservation methods on lung adenocarcinoma cell lines. Methods : Abandon the use of antibiotics in the culture medium, control cell passage and digestion conditions, and use rapid freezing method to preserve cells. RESULTS: At 37°C, 5% CO2 incubator began to adhere for about 2 h, completely adherent at 8 h, and monolayer at 48-60 h. Conclusions: (1) As long as the aseptic procedures are carefully observed, no routine use of antibiotics is required in the culture medium; (2) Control of trypsin conditions and centrifugation speed can reduce the damage during cell culture; (3) Cell freezing method is better than Traditional slow freezing method.
