目的:观察五色梅总黄酮对风湿性关节炎的治疗作用。方法:采用完全弗氏佐剂(CFA)诱导关节炎大鼠模型,设正常组,模型组,阳性药地塞米松组(0.15 mg·kg-1),五色梅总黄酮高、低剂量组(100,25 mg·kg-1)。ig给药各组每天1次,连续16 d,观察造模后第3,8,16,21,24天大鼠足跖肿胀率及关节炎指数,测定大鼠炎性组织液和血清中的白细胞介素-1β(IL-1β)及肿瘤坏死因子-α(TNF-α)的水平。小鼠二甲苯耳廓肿胀法和大鼠棉球肉芽肿法观察五色梅总黄酮的抗炎作用;小鼠热板法和醋酸扭体法观察五色梅总黄酮的镇痛作用。结果:与模型组比较,五色梅总黄酮100 mg·kg-1剂量组能降低佐剂性关节炎大鼠足肿胀率并降低关节炎指数(P<0.05),并能降低佐剂性关节炎大鼠炎性组织液和血清中IL-1β,TNF-α水平(P<0.05);五色梅总黄酮140,70 mg·kg-1剂量组均能显著抑制二甲苯所致小鼠耳廓肿胀程度,100 mg·kg-1剂量组能显著抑制大鼠棉球肉芽组织质量(P<0.05,P<0.01);五色梅总黄酮140,70 mg·kg-1剂量组均能显著减少醋酸引起小鼠的扭体反应次数和增加小鼠热板痛阈值(P<0.05)。结论:五色梅总黄酮有效改善类风湿性关节炎大鼠关节炎症状,同时具有显著的抗炎镇痛作用,其机制可能与降低炎性组织液和血清中IL-1β,TNF-α水平有关。
Objective: To observe the therapeutic effect of total flavones of Plum color on rheumatoid arthritis. Methods: The arthritis rats were induced by complete Freund’s adjuvant (CFA). The rats in normal group, model group, dexamethasone group (0.15 mg · kg-1), high and low dose group 100, 25 mg · kg-1). ig administration group once a day for 16 consecutive days to observe the rat’s plantar swelling rate and arthritis index on the 3rd, 8th, 16th, 21st and 24th days after modeling, and the levels of inflammatory cells and serum leukocytes Interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α) levels. Mouse xylene auricle swelling method and rat cotton ball granuloma were observed anti-inflammatory effect of the total flavonoids pentamers; mouse hot plate method and acetic acid writhing observed pentosial flavonoids analgesic effect. Results: Compared with the model group, 100 mg · kg -1 flavone total flavone decreased the paw edema and arthritis index (P <0.05) and decreased the rate of adjuvant arthritis The levels of IL-1β and TNF-α in the inflammatory tissue and serum of rats were significantly higher than those of the control group (P <0.05). The total flavonoids of Radix Aconiti Kusnezoffii in the doses of 140,70 mg · kg-1 could significantly inhibit the auricle swelling of mice induced by xylene , 100 mg · kg-1 dose group could significantly inhibit the granulation tissue quality of cotton ball (P <0.05, P <0.01); The number of writhing reactions in mice and the increase of the threshold of thermochromic pain in mice (P <0.05). CONCLUSION: The total flavones of P. plum can effectively reduce the symptoms of arthritis in rheumatoid arthritis rats, and have a significant anti-inflammatory and analgesic effect at the same time. The mechanism may be related to reducing the levels of IL-1β and TNF-α in inflammatory tissue and serum.
