目的探讨Claudin-4在胃癌中的表达及其临床病理意义。方法收集福建医科大学附属厦门第一医院2007年9月至2009年9月部分胃癌手术标本共68例。其中20例分别制成石蜡切片和留取新鲜冰冻标本,分别应用免疫组织化学染色方法和实时定量PCR方法检测该20例胃癌标本中Claudin-4蛋白和mRNA在胃癌组织、正常胃组织中的表达情况。另48例胃癌手术标本仅制成石蜡切片,采用免疫组织化学染色方法检测Claudin-4蛋白在胃癌组织中的表达情况。分析Clau-din-4表达情况与胃癌分化、浸润、转移和组织分型间的相关性。结果胃癌组织中Claudin-4的表达显著高于正常胃上皮组织(P<0.05);Claudin-4在Lauren分型肠型胃癌和BorrmannⅠ型、Ⅱ型胃癌组织中的表达分别显著高于弥漫型胃癌和BorrmannⅢ型、Ⅳ型胃癌(P<0.05);Claudin-4的表达与胃癌分化相关,胃癌分化越好,Claudin-4表达越高(P<0.05)。结论 Claudin-4与胃癌的发生、发展及胃癌的生物学行为密切相关,并可能成为预测胃癌生物学行为的重要指标。
Objective To investigate the expression of Claudin-4 in gastric carcinoma and its clinicopathological significance. Methods A total of 68 gastric cancer specimens collected from September 2007 to September 2009 in Xiamen First Hospital Affiliated to Fujian Medical University were collected. Twenty of them were made into paraffin sections and fresh frozen specimens respectively. The expression of Claudin-4 protein and mRNA in gastric cancer tissues and normal gastric tissues were detected by immunohistochemical staining and real-time quantitative PCR respectively Happening. The other 48 gastric cancer specimens were made into paraffin sections only. The expression of Claudin-4 protein in gastric cancer tissues was detected by immunohistochemical staining. The relationship between Clau-din-4 expression and differentiation, invasion, metastasis and histological type of gastric cancer was analyzed. Results The expression of Claudin-4 in gastric cancer tissues was significantly higher than that in normal gastric epithelium tissues (P <0.05). The expression of Claudin-4 in Lauren-typed intestinal-type gastric cancer and Borrmann typeⅠ and Ⅱ gastric cancer tissues was significantly higher than that in diffuse gastric cancer (P <0.05). The expression of Claudin-4 was correlated with the differentiation of gastric cancer. The better the differentiation of gastric cancer, the higher the expression of Claudin-4 (P <0.05). Conclusion Claudin-4 is closely related to the occurrence and development of gastric cancer and the biological behavior of gastric cancer, and may be an important index for predicting the biological behavior of gastric cancer.
