目的探讨盘状结构域受体2(DDR2)在肾透明细胞癌与正常肾组织中的表达及其临床意义。方法分别采用免疫组织化学染色和Real-time PCR检测DDR2在肾透明细胞癌及相应癌旁正常肾组织中的表达,进一步分析其表达与肾透明细胞癌相关临床病理特征间的关系。结果肾透明细胞癌组织中DDR2蛋白及mRNA与癌旁组织比较均呈现异常低表达(P<0.05),且肾透明细胞癌组织中DDR2的表达与肾癌临床分期及组织学分级呈负相关(P<0.05)。结论 DDR2在肾透明细胞癌组织中的表达较正常肾组织低,且其表达量与肾癌的临床分期和病理分级呈负相关,提示DDR2可能在肾透明细胞癌中扮演抑癌基因的角色,但具体机制还需进一步深入研究。
Objective To investigate the expression and clinical significance of discoid domain receptor 2 (DDR2) in renal clear cell carcinoma and normal renal tissue. Methods Immunohistochemical staining and Real-time PCR were used to detect the expression of DDR2 in renal clear cell carcinoma and its adjacent normal renal tissue respectively. The relationship between the expression of DDR2 and the clinicopathological features of renal clear cell carcinoma was further analyzed. Results The expression of DDR2 protein and mRNA in renal clear cell carcinoma tissues were significantly lower than that in adjacent noncancerous tissues (P <0.05). The expression of DDR2 in renal clear cell carcinoma was negatively correlated with the clinical stage and histological grade P <0.05). Conclusions The expression of DDR2 in renal clear cell carcinoma tissue is lower than that in normal renal tissue, and its expression level is negatively correlated with the clinical stage and pathological grade of renal cell carcinoma, suggesting that DDR2 may play a role of tumor suppressor gene in renal clear cell carcinoma, However, the specific mechanism needs further study.
