摘 要:目的 研究mdx鼠体内骨骼肌细胞凋亡情况,探讨凋亡对 Duchenne型肌营养不良症(DMD)的发生发展所起的作用。方法 采用流式细胞仪碘化丙啶(PI)排斥法测定不同渗透压下成肌细胞的凋亡数目,用TUNEL法测定不同鼠龄的mdx鼠骨骼肌细胞的凋亡情况。结果mdx鼠的成肌细胞在外界渗透压变化的情况下,发生凋亡的细胞数较C57鼠明显增加,且随mdx鼠鼠龄的增长而增加。结论 抗肌萎缩蛋白(dys)的缺失引起的凋亡在DMD疾病的启动和病程进展中可能起重要作用。
Abstract: Objective To investigate the apoptosis of skeletal muscle cells in mdx mice and to explore the role of apoptosis in the development of Duchenne muscular dystrophy (DMD). Methods Flow cytometry propidium iodide (PI) exclusion method was used to determine the number of myoblasts under different osmotic pressure. TUNEL method was used to determine the apoptosis of skeletal myocytes in different age groups. Results The number of apoptotic cells in mdx murine myoblasts increased significantly compared with that of C57 mice and increased with the increase of mice age. Conclusion Apoptosis induced by dystrophin dysfunction may play an important role in the initiation and progression of DMD.
