目的 研究乳腺癌间质新生血管周细胞形态学特点、特异性标记及其与内皮细胞的关系 ,并定量分析周细胞与血管密度的关系。方法 应用超微结构、免疫组织化学LSAB法及形态定量 ,对 89例乳腺癌及 4例新鲜肉芽组织新生血管的周细胞进行系统形态学观察。结果 内皮细胞第八因子相关抗原 (FⅧRAg)表达阳性 ,胞质内含有特征性的Ⅷ因子小体 (Weibel paladebody)。周细胞α 平滑肌肌动蛋白 (α SMA)表达阳性 ,胞质内含有丰富的肌丝 ,与内皮细胞间可见连接。癌间质内新生血管的周细胞与血管密度的比值 ,低血管密度区 (1.4± 0 3)明显高于高血管密度区 (4 .3± 0 .9) ,P <0 .0 0 0 5。结论 α SMA免疫组织化学染色结合部位和超微结构可作为研究周细胞的有用指标 ,周细胞在血管生成中可能起负向调节作用
Objective To study the morphological features, specific markers and their relationship with endothelial cells in pericancerous neovascularization of breast cancer, and quantitatively analyze the relationship between pericytes and vascular density. Methods Ultrastructural, immunohistochemical LSAB and morphometric quantification were used to observe the morphological changes of pericytes in 89 cases of breast cancer and 4 cases of fresh granulation tissue. Results The expression of FVIIIRAg in endothelial cells was positive, and the cytoplasm contained characteristic Weibel palade bodies. Peripheral α smooth muscle actin (α SMA) expression is positive, the cytoplasm is rich in myofilament, visible junctions with endothelial cells. The ratio of pericytes to vascular density in neovascularization in cancer interstitium was significantly higher in the low vascular density area (1.4 ± 0.3) than in the high vascular density area (4.3 ± 0.9). P <0 0 0 0 5 . Conclusion The binding site and ultrastructure of α SMA immunohistochemical staining can be used as a useful indicator for the study of pericytes. Pericytes may play a negative role in angiogenesis.
