目的 临床上发现用可乐或牛奶送服伊曲康唑胶囊可提高其生物利用度和治疗效果,本研究旨在比较伊曲康唑胶囊微丸在蒸馏水、牛奶(全脂、低脂以及脱脂牛奶,伊利乳业)和可乐(可口可乐)中的溶出度.方法 进行伊曲康唑微丸溶出度试验,采用液相色谱-串联质谱法对伊曲康唑微丸在溶出度试验中不同阶段的溶出浓度进行检测.结果 自然pH的蒸馏水、牛奶、可乐中,伊曲康唑溶出度:可乐>牛奶>蒸馏水.从溶解平均最高浓度来看,可乐溶解伊曲康唑的能力约是牛奶的2.36 ~3.19倍,是蒸馏水的10.37倍.在蛋白质含量相同的情况下,全脂牛奶(脂肪6%)在牛奶中溶出能力最强.结论 推荐临床上使用可乐或者全脂牛奶送服伊曲康唑胶囊,可增加药物溶出度,促进胃肠道吸收.“,”Objective In the experience of clinicians,the bioavailability and the therapeutic effect of itraconazole capsules can be evaluated by taking with liquids such as cola or milk.In this study,we aimed to compare the dissolution of itraconazole capsule pellets in distilled water,milk (whole-fat,low-fat and skimmed milk,Yi-Li) and cola (Coca-Cola).Methods The concentration of dissolution medium in the dissolution test of itraconazole pellets was detected by liquid chromatography-tandem mass spectrometry.Results For the normal pH of distilled water,milk and cola,the dissolubility of itraconazole pellets was cola > milk > distilled water.At the highest average concentration of dissolution,the dissolubility of itraconazole in cola was about 2.36 to 3.19 times of milk and 10.37 times of distilled water.Among the three kinds of milk,with the same protein content,the whole-fat milk (fat 6%) was the strongest dissolving medium.Conclusion It is recommended that itraconazole capsule is taken with cola or whole-fat milk,for they both can enhance the dissolubility of itraconazole and promote the absorption of the drug.
