目的探讨中国广东汉族人群葡萄糖转运蛋白-4(GLUT4)基因多态性与冠心病易感性之间的关系。方法采用多重单核苷酸多态性(SNP)分型试剂盒,对筛选的2个tag SNP位点(rs16956647、rs5435)在1 044例冠心病患者和1 349例非冠心病患者为对照进行基因分型;采用遗传分析软件Plink和统计分析软件SPSS 17.0分析SNP位点与冠心病的关联性。结果 SNP rs5435未满足Hardy-Weinberg平衡条件,予以剔除。校正性别、年龄、高血压、糖尿病、吸烟史、总胆固醇、甘油三酯、心率和体质指数后,rs16956647基因型与冠心病存在明显关联[加性模型:P=0.016,OR=0.80(95%CI=0.67~0.96);显性模型:P=0.002,OR=0.71(95%CI=0.57~0.89)]。进一步按是否患糖尿病分层后进行基因型关联分析,结果显示仅在糖尿病组rs16956647位点与冠心病呈显著关联[加性模型:P=0.048,OR=0.82(95%CI=0.67~0.99);显性模型:P=0.007,OR=0.72(95%CI=0.56~0.91)]。结论在中国广东汉族人群中,GLUT4基因多态性与冠心病易感性之间的关联可能与患者胰岛素抵抗有关。
Objective To investigate the relationship between GLUT4 gene polymorphism and susceptibility to coronary heart disease in Han nationality in Guangdong province, China. Methods Two SNPs (rs16956647, rs5435) were screened out in 1 044 patients with coronary heart disease and 1 349 patients with non-coronary heart disease as controls, using multiplex single nucleotide polymorphism (SNP) typing kit. Genotypes were analyzed using genetic analysis software Plink and statistical analysis software SPSS 17.0 to analyze the association between SNP loci and coronary heart disease. Results SNP rs5435 did not meet the Hardy-Weinberg equilibrium conditions, be removed. The rs16956647 genotype was significantly associated with coronary heart disease after adjusting for gender, age, hypertension, diabetes, smoking history, total cholesterol, triglyceride, heart rate and body mass index [additive model: P = 0.016, OR = CI = 0.67-0.96); Dominant model: P = 0.002, OR = 0.71 (95% CI = 0.57-0.89)]. Further analysis of genotypes showed that there was a significant association between rs16956647 and coronary heart disease in the diabetic group [additive model: P = 0.048, OR = 0.82 (95% CI = 0.67-0.99) ; Dominant model: P = 0.007, OR = 0.72 (95% CI = 0.56-0.91)]. Conclusion The association between GLUT4 gene polymorphism and susceptibility to coronary heart disease may be related to insulin resistance in Chinese Han population of Guangdong province.
