目的 观察钙离子拮抗剂尼莫地平 ( NMD)与化疗药物卡氮介 ( BCNU)及威猛 ( Vm-2 6)联合使用对脑胶质瘤细胞化疗耐药性的影响 ,为临床上脑胶质瘤的合理化疗提供理论依据。方法 通过体外培养的 C6胶质瘤细胞 ,采用 MTT法及 3 H-Td R掺入法 ,比较 NMD与 BCNU、Vm-2 6单独使用、联合使用对肿瘤细胞增殖率的影响。结果 NMD分别与 BCNU、Vm-2 6联合使用对肿瘤细胞的抑制率明显高于三者单独使用的抑瘤效果 ( P<0 .0 1 ) ,而三者联合使用对肿瘤细胞的抑制率可达 97.0 4 % ,也明显高于 NMD与 BCNU或 Vm-2 6联合使用的抑瘤率 ( P <0 .0 1 )。同时 ,联合化疗时 BCNU、Vm-2 6的用药浓度仅为单独使用的 0 .1~ 0 .0 1。结论 NMD可以明显降低胶质瘤细胞对化疗药物 BCNU和 Vm-2 6的耐药性 ,而且可以降低化疗药物的用药浓度。
Objective To observe the effect of combination of calcium antagonist nimodipine (NMD) and chemotherapeutic agents BCNU and Vm-2 6 on chemoresistance in glioma cells, Reasonable chemotherapy of tumor provides the theoretical basis. Methods The C6 glioma cells cultured in vitro were treated with MTT and 3 H-Td R incorporation to compare the effects of NMD with BCNU and Vm-2 6 alone or in combination on the proliferation of tumor cells. Results The inhibitory rates of NMD combined with BCNU and Vm-2 6 on tumor cells were significantly higher than those of the other three alone (P <0.01), and the inhibitory rates of NMD combined with BCNU and Vm-2 6 on tumor cells were Up to 97.0 4%, which was also significantly higher than that of NMD combined with BCNU or Vm-2 6 (P <0.01). At the same time, BCNU, Vm-2 6 drug concentration when combined chemotherapy only used alone 0.1 ~ 0. Conclusion NMD can significantly reduce the drug resistance of glioma cells to chemotherapy drugs BCNU and Vm-2 6, and can reduce the concentration of chemotherapy drugs.
