目的:探讨葛根素对糖尿病大鼠心肌的形态结构、功能及NF-κB表达的影响。方法:以45mg/kg剂量一次性尾静脉注射链脲佐菌素建立SD大鼠实验性糖尿病模型。应用葛根素(40mg/kg.d、80mg/kg.d)腹腔注射治疗8周后,测定各组大鼠血糖、血清LDH、CK含量及心肌组织SOD活性、MDA含量,检测心功能,应用光镜、电镜观察大鼠心肌形态结构,免疫组化方法观察心肌组织NF-κB表达情况。结果:与糖尿病模型组比较,葛根素治疗组大鼠血糖水平稍降,血清LDH、CK含量及心肌组织MDA含量显著降低,而高剂量组SOD活性明显增强,左心室功能、心肌形态结构尤其是超微结构明显改善,心肌组织NF-κB阳性表达明显减少。结论:葛根素对糖尿病大鼠心肌具有一定的保护作用,能明显改善心功能和心肌超微结构,可能与其减弱糖尿病大鼠心肌过氧化损伤及减少心肌组织NF-κB阳性表达有关。
Objective: To investigate the effect of puerarin on myocardium morphological structure, function and NF-κB expression in diabetic rats. METHODS: SD rats were injected with streptozotocin at a dose of 45 mg/kg to establish an experimental diabetic model. After puerarin (40mg/kg.d, 80mg/kg.d) was intraperitoneally injected for 8 weeks, blood glucose, serum LDH, CK content, SOD activity and MDA content in myocardial tissue were measured, and cardiac function was detected. The morphology and structure of myocardial tissue were observed by microscope and electron microscope. The expression of NF-κB in myocardial tissue was observed by immunohistochemistry. RESULTS: Compared with the diabetic model group, puerarin treatment group rats blood glucose levels decreased, serum LDH, CK content and myocardial tissue MDA content decreased significantly, and high dose group SOD activity was significantly enhanced, left ventricular function, myocardial morphology and structure in particular The ultrastructure was significantly improved and the positive expression of NF-κB in myocardial tissue was significantly reduced. Conclusion: Puerarin has a certain protective effect on the myocardium of diabetic rats, and can significantly improve the cardiac function and myocardial ultrastructure. It may be related to the weakening of myocardial peroxidation injury and the decrease of myocardial NF-κB expression in diabetic rats.
