目的:探讨柑橘皮总黄酮(TFO)对大鼠佐剂性关节炎(AA)发展的影响及机制。方法:建立大鼠AA模型,灌胃给药37.5,75,150 mg.kg-1TFO。游标卡尺测量大鼠足跖厚度,HE染色观察关节病理损伤,放射免疫法检测血清肿瘤坏死因子(TNF)-α、白介素(IL)-1β和前列腺素(PG)E2水平,Western blot法分析关节滑膜组织环加氧酶(COX)-2表达。结果:75,150 mg.kg-1TFO可显著减少AA大鼠足跖厚度,减轻关节病理学损伤,降低血清TNF-α,IL-1β,PGE2水平,抑制关节滑膜组织COX-2表达。结论:TFO可拮抗大鼠AA发展,其作用机制与抑制炎症介质的生成密切相关。
Objective: To investigate the effect and mechanism of TFO on the development of adjuvant arthritis (AA) in rats. Methods: The rat model of AA was established and intragastrically administrated 37.5, 75, 150 mg.kg-1TFO. The thickness of the plantar paw was measured by vernier calipers, the pathological damage of joints was observed by HE staining, the levels of tumor necrosis factor (TNF) -alpha, interleukin (IL) -1β and prostaglandin (PG) E2 were detected by radioimmunoassay Membrane tissue cyclooxygenase (COX) -2 expression. Results: 75,150 mg.kg-1TFO significantly reduced the thickness of plantar paws in AA rats, reduced the pathological injury of joints, decreased the levels of TNF-α, IL-1β and PGE2, and inhibited the expression of COX-2 in synovial tissues. Conclusion: TFO can antagonize the development of rat AA and its mechanism of action is closely related to the inhibition of inflammatory mediators.
