目的观察天麻钩藤饮对血管紧张素Ⅱ(AngⅡ)诱导血管平滑肌细胞(A7r5)增殖的影响及机制。方法 AngII刺激A7r5细胞建立细胞增殖模型。采用CCK-8法检测细胞增殖,观察天麻钩藤饮(0.25,0.5,1,2 mg.mL-1)、亚硝基-精氨酸甲酯(100μmo1.L-1)对AngII诱导血管平滑肌细胞增殖的影响。硝酸还原酶及化学比色法测定细胞上清液中一氧化氮(NO)、一氧化氮合酶(NOS)、诱导型一氧化氮合酶(iNOS)水平。结果天麻钩藤饮在0.25~2 mg.mL-1呈剂量及时间依赖性抑制A7r5增殖;天麻钩藤饮能升高NO、NOS、iNOS水平。结论天麻钩藤饮对AngII诱导的A7r5细胞增殖有抑制作用,升高NO和NOS水平可能是其发挥作用的机制。
Objective To observe the effect and mechanism of Tianma Gouteng Yin on the proliferation of vascular smooth muscle cells (A7r5) induced by angiotensin Ⅱ (AngⅡ). Methods AngII stimulated A7r5 cells to establish cell proliferation model. Cell proliferation was measured by CCK-8 assay. The effects of Tianma Gouteng Decoction (0.25,0.5,1,2 mg.mL-1) and nitroso-arginine methyl ester (100μmo1.L-1) on Ang II-induced vascular smooth muscle Effect of cell proliferation. Nitric acid reductase and chemical colorimetry were used to determine the level of nitric oxide (NO), nitric oxide synthase (NOS) and inducible nitric oxide synthase (iNOS) in the cell supernatant. Results Tianma Gouteng Decoction inhibited the proliferation of A7r5 in a dose-and time-dependent manner at a dosage of 0.25-2 mg.mL-1. Tianma Gouteng Decoction could increase NO, NOS and iNOS levels. Conclusion Tianma Gouteng Decoction can inhibit the proliferation of A7r5 cells induced by AngII. Increasing the levels of NO and NOS may play a role in the mechanism.
